    Exploring the effect of sex on empirical fitness landscapes

    The nature of epistasis has important consequences for the evolutionary significance of sex and recombination. Recent efforts to find negative epistasis as a source of negative linkage disequilibrium and associated long-term advantage to sex have yielded little support. Sign epistasis, where the sign of the fitness effects of alleles varies across genetic backgrounds, is responsible for the ruggedness of the fitness landscape, with several unexplored implications for the evolution of sex. Here, we describe fitness landscapes for two sets of strains of the asexual fungus Aspergillus niger involving all combinations of five mutations. We find that 30% of the single-mutation fitness effects are positive despite their negative effect in the wild-type strain and that several local fitness maxima and minima are present. We then compare adaptation of sexual and asexual populations on these empirical fitness landscapes by using simulations. The results show a general disadvantage of sex on these rugged landscapes, caused by the breakdown by recombination of genotypes on fitness peaks. Sex facilitates movement to the global peak only for some parameter values on one landscape, indicating its dependence on the landscape’s topography. We discuss possible reasons for the discrepancy between our results and the reports of faster adaptation of sexual population

    EXPERIMENTAL AND COMPUTATIONAL ACTIVITIES AT THE OREGON STATE UNIVERSITY NEES TSUNAMI RESEARCH FACILITY

    A diverse series of research projects have taken place or are underway at the NEES Tsunami Research Facility at Oregon State University. Projects range from the simulation of the processes and effects of tsunamis generated by sub-aerial and submarine landslides (NEESR, Georgia Tech.), model comparisons of tsunami wave effects on bottom profiles and scouring (NEESR, Princeton University), model comparisons of wave induced motions on rigid and free bodies (Shared-Use, Cornell), numerical model simulations and testing of breaking waves and inundation over topography (NEESR, TAMU), structural testing and development of standards for tsunami engineering and design (NEESR, University of Hawaii), and wave loads on coastal bridge structures (non-NEES), to upgrading the two-dimensional wave generator of the Large Wave Flume. A NEESR payload project (Colorado State University) was undertaken that seeks to improve the understanding of the stresses from wave loading and run-up on residential structures. Advanced computational tools for coupling fluid-structure interaction including turbulence, contact and impact are being developed to assist with the design of experiments and complement parametric studies. These projects will contribute towards understanding the physical processes that occur during earthquake generated tsunamis including structural stress, debris flow and scour, inundation and overland flow, and landslide generated tsunamis. Analytical and numerical model development and comparisons with the experimental results give engineers additional predictive tools to assist in the development of robust structures as well as identification of hazard zones and formulation of hazard plans

    The in vitro effects of dehydroepiandrosterone on human osteoarthritic chondrocytes

    AbstractObjective: To investigate the in vitro effects of dehydroepiandrosterone (DHEA) on human osteoarthritic chondrocytes.Design: Chondrocytes isolated from human osteoarthritic knee cartilage were three-dimensionally cultured in alginate beads, except for cell proliferation experiment. Cells were treated with DHEA in the presence or absence of IL-1β. The effects on chondrocytes were analyzed using a 3-(4,5-dimethylthiazol-2yl)-5-(3-carboxymethoxy-phenyl)-2-(4-sulfophenyl)-2H-tetrazolium inner salt (MTS) assay (for chondrocyte proliferation), a dimethylmethylene blue (DMB) assay (for glycosaminoglycan (GAG) synthesis), and an indole assay (for DNA amount). Gene expressions of type I and II collagen, metalloproteinase-1 and -3 (MMP-1 and -3), and tissue inhibitor of metalloproteinase-1 (TIMP-1) as well as the IL-1β-induced gene expressions of MMP-1 and -3 were analyzed by reverse transcription-polymerase chain reaction (RT-PCR). The protein synthesis of MMP-1 and -3 and TIMP-1 was determined by Western blotting.Results: The treatment of chondrocytes with DHEA did not affect chondrocyte proliferation or GAG synthesis up to 100μM of concentration. The gene expression of type II collagen increased in a dose-dependent manner, while that of type I decreased. DHEA suppressed the expression of MMP-1 significantly at concentrations exceeding 50μM. The gene expression of MMP-3 was also suppressed, but this was without statistical significance. The expression of TIMP-1 was significantly increased by DHEA at concentrations exceeding 10μM. The effects of DHEA on the gene expressions of MMP-1 and -3 were more prominent in the presence of IL-1β, in which DHEA suppressed not only MMP-1, but also MMP-3 at the lower concentrations, 10 and 50μM, respectively. Western blotting results were in agreement with RT-PCR, which indicates that DHEA acts at the gene transcription level.Conclusions: Our study demonstrates that DHEA has no toxic effect on chondrocytes up to 100μM of concentration and has an ability to modulate the imbalance between MMPs and TIMP-1 during OA at the transcription level, which suggest that it has a protective role against articular cartilage loss

    Extremely large perpendicular magnetic anisotropy of an Fe(001) surface capped by 5d transition metal monolayers: A density functional study

    Significant enhancement of the magnetocrystalline anisotropy (MCA) of an Fe(001) surface capped by 4d and 5d transition metal monolayers is presented in this study using first principles density functional calculations. In particular, an extremely large perpendicular MCA of +10 meV/Ir was found in Ir-capped Fe(001), which originates not from the Fe but from the large spin-orbit coupling of the Ir atoms. From the spin-channel decomposition of the MCA matrix and electronic structure analyses, we find that strong 3d-5d band hybridization in the minority spin state is responsible for the sign changes of the MCA from parallel to perpendicular.open0

    Spintronic properties of one-dimensional electron gas in graphene armchair ribbons

    We have investigated, using effective mass approach (EMA), magnetic properties of a one-dimensional electron gas in graphene armchair ribbons when the electrons of occupy only the lowest conduction subband. We find that magnetic properties of the one-dimensional electron gas may depend sensitively on the width of the ribbon. For ribbon widths Lx=3Ma0L_x=3Ma_0, a critical point separates ferromagnetic and paramagnetic states while for Lx=(3M+1)a0L_x=(3M+1)a_0 paramagnetic state is stable (MM is an integer and a0a_{0} is the length of the unit cell). These width-dependent properties are a consequence of eigenstates that have a subtle width-dependent mixture of K\mathbf{K} and K\mathbf{K'} states, and can be understood by examining the wavefunction overlap that appears in the expression for the many-body exchange self-energy. Ferromagnetic and paramagnetic states may be used for spintronic purposes.Comment: 5 pages, 6 figure

    Non-minimally coupled scalar field cosmology on the phase plane

    In this publication we investigate dynamics of a flat FRW cosmological model with a non-minimally coupled scalar field with the coupling term ξRψ2\xi R \psi^{2} in the scalar field action. The quadratic potential function V(ψ)ψ2V(\psi)\propto \psi^{2} is assumed. All the evolutional paths are visualized and classified in the phase plane, at which the parameter of non-minimal coupling ξ\xi plays the role of a control parameter. The fragility of global dynamics with respect to changes of the coupling constant is studied in details. We find that the future big rip singularity appearing in the phantom scalar field cosmological models can be avoided due to non-minimal coupling constant effects.     Z boson pair production at LHC in a stabilized Randall-Sundrum scenario

    We study the Z boson pair production at LHC in the Randall-Sundrum scenario with the Goldberger-Wise stabilization mechanism. It is shown that comprehensive account of the Kaluza-Klein graviton and radion effects is crucial to probe the model: The KK graviton effects enhance the cross section of ggZZg g \to Z Z on the whole so that the resonance peak of the radion becomes easy to detect, whereas the RS effects on the qqˉZZq\bar{q} \to Z Z process are rather insignificant. The pTp_T and invariant-mass distributions are presented to study the dependence of the RS model parameters. The production of longitudinally polarized Z bosons, to which the SM contributions are suppressed, is mainly due to KK gravitons and the radion, providing one of the most robust methods to signal the RS effects. The 1σ1 \sigma sensitivity bounds on (Λπ,mϕ)(\Lambda_\pi, m_\phi) with k/MPl=0.1k/M_{\rm Pl} =0.1 are also obtained such that the effective weak scale Λπ\Lambda_\pi of order 5 TeV can be experimentally probed.Comment: 28 pages, LaTex file, 18 eps figure
